HEK293 clones expressing NOD1

293/NOD1 cells were obtained by stable transfection of HEK293 cells with the pUNO-hNOD1 plasmid which expresses the human NOD1 gene or with the pUNO-mNOD1 plasmid which expresses the murine NOD1 gene.

The control cell line for 293/NOD1 cells is 293-null cells.

293/TLR, NOD and control clones were transfected transiently with pNiFty-SEAP and stimulated with TLR and NOD ligands. After 16 hour stimulation, NF-κB-induced SEAP activity was assessed using QUANTI-Blue™, a SEAP detection medium.

293/hNOD1 cells transiently transfected with the NF-κB-inducible SEAP reporter plasmid, pNiFty-SEAP, were incubated with the NOD1 ligands, iE-DAP, Tri-DAP, and C12-iE-DAP, and the NOD2 ligands, MDP and murabutide, at various concentrations. After 24h incubation, NOD1 stimulation was assessed by measuring the levels of SEAP secreted in the supernatant by using HEK-Blue™ Detection medium, which turns blue in the presence of SEAP.
